What a "black box warning" really means

A black box warning is the FDA's strongest prescription-label warning. It means there's a serious or life-threatening risk worth highlighting — not that the medication shouldn't be used, but that providers and patients should be aware of specific precautions.

GLP-1 medications and thyroid C-cell tumors

The branded GLP-1 medications (Wegovy®, Ozempic®, Zepbound®, Mounjaro®) carry a black box warning regarding thyroid C-cell tumors, based on rodent studies. These medications are contraindicated for patients with a personal or family history of medullary thyroid carcinoma (MTC) or Multiple Endocrine Neoplasia syndrome type 2 (MEN 2).
